Effects of Daphnetin on Experimental Acute Pancreatitis-Associated Acute Lung Injury in Mice

Background and Aim. Daphnetin, an active monomer ingredient extracted from D. marginata, is proved to have anti-inflammatory antioxidant effect. The aim of this study explore the effect possible mechanism daphnetin on acute lung injury (ALI) associated with pancreatitis (AP) in mice. Methods. A total 36 mice were randomly assigned into three groups: control group, AP group. mouse model was induced by caerulein lipopolysaccharide. Animals sacrificed at 6 12 h after treatment, respectively. pathological changes pancreas determined hematoxylin-eosin staining scores. Levels IL-1β, IL-6, TNF-α serum activity myeloperoxidase (MPO) tissue homogenate detected ELISA. protein level toll-like receptor 4 (TLR4), phospho-nuclear factor-kappa B p65 (p-NF-κB p65), nuclear (NF-κB hypoxia-inducible factor 1 alpha (HIF-1α) Western blot. Results. Results showed extensive neutrophil infiltration, hemorrhage, edema tissues or AP. treatment improved MPO levels inflammatory cytokines including TNF-α, IL-6 group significantly higher than those ( P < 0.05), intervention reversed id="M2"> 0.05). Compared mice, TLR4, p-NF-κB p65, HIF-1α id="M3"> while decreased these expression levels. No significant difference observed NF-κB among control, AP, groups id="M4"> > Conclusions. Daphnetin exerted a protective SAP may be related regulation TLR4/NF-κB/HIF-1α pathway reduce release factors.
